苯丙素苷类化合物的研究进展 (Ⅰ)

       苯丙素苷化合物是一类含有苯乙基和取代肉桂酰基的天然糖苷,因其具有广泛多样的生理活性而日益受到人们的重视。该文对近十年来新分离出的苯丙素苷类化合物做一总结。

       Abstract: Phenylpropanoid Glycosides (PPGs) are natural glycosides exist widely in folk herbal plants. A rapid increased isolation in the number of PPGs has been reviewed in this article in recent ten years.

        苯丙素苷化合物 (phenylpropanoid glycosides,PPGs) 是一类含取代苯乙基和肉桂酰基的天然糖苷,广泛存在于双子叶植物中。通常是以βD吡喃葡萄糖为中心糖核,取代苯乙基为其苷元,取代肉桂酰基以酯键与糖核相连,中心糖核的C6,C3位上还可以连接其它的糖。因其苷元为取代苯乙基,亦称为苯乙醇苷。由于糖的种类的变化,支链糖的连接位置、顺序的不同,以及肉桂酰基和苯乙基上取代基团的变化,从而导致了该类化合物结构类型的多样性。近年的研究表明,该类化合物作为药用植物的主要有效成分具有广泛而显著的生理活性,因而日益受到植物化学家、药物学家及有机化学家的重视。本文对近十年来在苯丙素苷类化合物的提取分离方面的研究进展做一综述。
        苯丙素苷类化合物广泛存在于双子叶植物中, 自1963年第1个PPGs被分离以来,该类化合物数目不断增加,每年都有一些新的苯丙素苷化合物被分离、鉴定,到1993年其分离的总数目已达109个[1]。在近10年中又从近40种植物中分离鉴定了近80种苯丙素苷类化合物。见表1[2~40]。表1  已分离出的苯丙素苷化合物的名称和植物来源(略)
        苯丙素苷化合物可用80%乙醇,水,甲醇,CHCl3等初提,紫外(254,210 nm)或TLC定性检测。提取液减压浓缩后得悬浮液,然后用CH2Cl2,CHCl3, EtOAc, 和n-BuOH等萃取,之后过Sephadex G15或LH-20 柱(MeOH±H2O 1∶3 to 2∶1,或EtOH/H2O 1∶1,或CHCl3 /MeOH 2∶1),硅胶柱(正己烷/ EtOAc/Me2CO /MeOH, 石油醚/EtOAc), CC柱(EtOAc/MeOH/H2O)或 ODS 柱 (30% MeOH),flash chromatography (CHCl3 /Me2CO/ H2O)进行分离,上液相可用MeCN/H2O洗脱,进行定量检测。
        随着更多新的PPGs的分离和鉴定,糖类药物作为新一代的抗肿瘤药物将日趋受到人们的青睐与重视。(未完待续)
